Dr Rout was graduated (BVSc & AH) in 1987 in First Division with 72.8% marks from the College of Veterinary Science and Animal Husbandry (Orissa University of Agriculture & Technology), Bhubaneswar, and Post-Graduation (MVSc) in Animal Genetics and Breeding in 1990, with 3.269/4.0 OGPA (First Division), and Doctoral Degree (PhD) in Animal Genetics and Breeding from the Indian Veterinary Research Institute (IVRI), Izatnagar, Uttar Pradesh in 1995 with 8.27/10 OGPA. He had worked as post-doctoral fellow for 2 years at National Institute of Immunology, New Delhi in Molecular Virology/ Molecular Biology at the National Institute of Immunology, New Delhi.
