Infertility suspected extractable and leachable study of phthalates in pharmaceutical products by TLC versus LC-MS/MS

Abstract :

Infertility suspected phthalates are used significantly in the making of plastic. These plastics are used in the pharmaceutical packagings. A simple and rapid method for simultaneous estimation of four phthalates, namely Dimethyl Phthalates (DMP), Diethyl Phthalates (DEP), Dibutyl Phthalates (DBP) and di-(2-Ethylhexyl) Phthalates (DEHP) in pharmaceutical products by HPTLC was developed. These phthalates suspected to irreversible infertility to human. The plastic pharmaceutical packaging material and content were extracted by liquid-liquid extraction. The filtered organic liquid was applied and the chromatographic separation was carried out on TLC aluminium foils silica gel 60 with n-hexane-ethyl acetate 9:1. Densitometric detection was performed at 240 nm. The linear regression of the calibration plots in the same concentration range of 100-1400 ng/band showed good correlation coefficients of r=0.9980, 0.9981, 0.9965 and 0.9962 for DMP, DEP, DBP and DEHP, respectively. The method was validated with respect to linearity, accuracy, precision and specificity. The several different pharmaceutical products were processed for the extractable and leachable study using the new method, which is shown to be the suitable for routine use for quality control and regulatory purpose. The proposed HPTLC method was compared with inhouse developed LC-MS/MS method in several parameters and HPTLC turns out to be the method of choice for a fast and economic analysis.

Biography :

Kashyap Thummar has his expertise in handling various sophisticated analytical instruments mainly HPTLC, GC-MS/MS and LC-MS/MS. He also extended his works on analytical work to find out prevalence of phthalates metabolite in human and their relation to established male infertility. His research interests include therapeutic drug monitoring, impurity profiling and bioautography.
