Mediforum developed a HPV screening kit (MF HPV HR screening) using the cocktails of digoxigenin-labeled human papillomavirus (HPV) type-specific oligonucleotide probes and an enzyme immunoassay (EIA) was used as a basis to develop a group-specific detection method for 15 high-risk HPV (types: 16, 18, 31, 33, 35, 39, 45, 51, 52, 56, 58, 59, 66 and 68,70), following a general primer MY11/bioGP6(+)-mediated PCR. We aimed to compare MF HPV HR screen with Qiagen HCII for evaluation and validation of MF HPV HR screening kit. Cervical scraps (specimens) were obtained from A-Ju medical Center and such specimens had previously gone through a liquid-based cytology and HPV screening by Qiagen HCII. 413 out of 455 specimens demonstrated the results as to the presence of HR-HPV in between the HPV HR kit and HCII (246 positive and 167 negative, 90.8%). The discordant results demonstrated in 42 specimens in between HPV HR and HCII (9.2%). For the overall analysis, MF HPV-HR assay kit demonstrated 96.1% of sensitivity, 95.8% of specificity, 96.1% of PPV and 97.1% of NPV. HCII assay kit demonstrated 89.3% of sensitivity, 95.8% of specificity, 95.7% of PPV and 89.7% of NPV. The result of MF HPV-HR assay showed higher sensitivity than HCII on H-SIL and L-SIL specimens, degree of concordance between MF HPV-HR and DNA sequencing assay was higher than that of between HCII and DNA sequencing assay. MF HPV-HR HPV assay would be effective to screen more common high risk HPV.
