Three new pterocarpans, velucarpins A-C and two new isoflavanes, dalvelutinanes A-B along with six known compounds were isolated from the roots of Dalbergia velutina. Their chemical structures including absolute configurations were determined by spectroscopic analysis (1D and 2D NMR, HRESIMS and CD). All isolated compounds were evaluated for their cytotoxicity against five cancer cell lines (KB, HeLa S-3, MCF-7, HepG-2 and HT-29). Compound 10 showed good cytotoxicity against all the five cancer cell lines with IC50 values in the range of 3.47-9.76 ├?┬ÁM. In addition, compound 3, 7 and 9 showed good cytotoxicity against KB and HeLa S-3 cells with IC50 values in the range of 5.99-9.54 ├?┬ÁM and compound 5 showed good cytotoxicity against MCF-7 cell with IC50 value of 4.69 ├?┬ÁM.
