Abstract :

Background and Purpose: Nephroblastoma is the most common pediatric tumor of the kidney. Wilms tumor patients are too young, can not clearly and accurately describe uncomfortable feeling, and the early clinical symptom is unclear. Protein biomarkers were identified in previous studies, but they were not checked out by long time and stability of expression level. The purpose of this study was to check out stability of expression level of protein biomarkers which were identified in previous studies. Materials and Methods: The total of 25 cells samples (divided into 50 parts) of pre-surgery were cultured in two weeks and six weeks. Different culture times of cells were divided into two groups. Two groups of cells homogenates were made from cells samples. The expression level of protein biomarkers was identified by comparison between groups using proteomics technologies. The study of protein biomarkers was the same as protein biomarkers which were in previous studies (11526 Da and 4756 Da). Results: In proteomics technologies and SELDI-TOF-MS, the expression level of protein is represented by peak value. Compared with the peak values of two peaks (11526 Da and 4756 Da), there was no statistical significance between group of cultured in two weeks and group of cultured in six weeks. Discussion: In the study, the expression level of biomarkers (11526 Da and 4756 Da) which were identified in previous studies has kept steady. The expression level of biomarkers was almost the same at different times and different stages. As time went on, the expression of biomarkers was not enhancing and reducing. According to this study, we demonstrated that tumor secreted protein without any external disturbance. In next step, we attempted to use the biomarkers to screened wilms tumor.
